Very simple, send a letter attaching a job description o he higher post. Obviously, the JD would contain the roles and responsibilities and also the minimum qualification required.

Dear Madam,

Promotion is management's prerogative, and it is not up to the employee to demand it. Please call her into your office and verbally convey this, as well as the fact that she does not possess the requisite qualifications and does not meet other conditions.
