LOI is nothing but Offer of employment letter which will generally be given to a candidate after successful completion of Interview, Pre employment medical check up. But the Appointment Letter will be issued after joining the job and sometimes after confirmation on completion of Probationary period.

It is like Provisional course completion certificate. Once Degree certificate is received, provisional certificate becomes invalid.
LOI - Letter of Intent is issued preceding the appointment letter.
