I joined a company on 27 Jan 2018. I got transferred in May 2019 to my hometown branch (transfer was requested by me), but that branch opened in Feb 2020.

Now, as per my joining date, I will complete 4 years and 240 calendar days in Sep 2022.

Can I claim gratuity if I leave the organization in Oct 2022 or Nov 2022? Or will the period from June 2019 to Jan 2020 be counted as 0?

Since you were allowed to leave without salary, the company does not treat it as a break in employment. However, since you did not work during that period, it will not count in the number of days for which you will be paid the gratuity.
