The moment that the Maternity Benefit Act,1961 becomes applicable to your establishment as per Sec. 2(1) of the Act, the first step you have to take is start maintaining the registers, notices etc., in the proforma mentioned in the Central or State Rules applicable and the second one is to abide by the provisions as and when any claim for maternity benefit is made.
There is no registration formalities under The Maternity Benefit Act. Only thing is you have to maintain certain registers in the prescribed format.
In addition, by virtue of Amendment in 2017, you have to comply with the following conditions

In addition, by virtue of amendment namely: The Maternity Benefit (Amendment) Act, 2017 you have to comply with the following conditions stipulated therein.
‘‘11A. (1) Every establishment having fifty or more employees shall have the
facility of créche within such distance as may be prescribed, either separately or alongwith common facilities :
Provided that the employer shall allow four visits a day to the creche by the
woman, which shall also include the interval for rest allowed to her.
(2) Every establishment shall intimate in writing and electronically to every
woman at the time of her initial appointment regarding every benefit available under
