I want to know whether spouse is covered whether he/she is working in another institution earning above 5000/-. As per ESIC rules Parents (dependents) who are having income more then 5000/- are not covered. Is same applicable to Spouse for Medical Benefit.

The income of the spouse is not a factor for deciding the ESI coverage and she is eligible irrespective of her income. As regards parents, Section 2 (12) (v) of the Act specifies an income ceiling to be eligible for the benefits under the Act while there is no such mention for spouse.
