whether the resignation letter sent by e mail without signature is acceptable legally. Normally , every organisation insist for handwritten resignation letter from the employees.

As per my knowledge is acceptable if the mail has come from his official email id. Moreover receiving resignation email from the employee is the strong proof for future verification.

You need to refer to Sec 4 of the information technology act and other relevant parts of the act. There is a clear provision that email is now equivalent to write communication. So resignation sent by email is valid, there is no need for signature.
The resignation need not be thorough official email id. It can very well be from a personal ID. in fact, the resignation from official I'd is a bad idea since at a later date, the employee will not have access to it.

Well… As per my current employer the resignation mails shall be considered in 02 different ways. We are receiving resignation alert through email and the employees are sending mails from their official email. Because most of the times they are in site and they are not getting a chance to meet the concern person to submit the resignation in written.
Receiving mail from personal email id shall be considered if the employee is out of station or on vacation. Hence submitting resignation through email shall be considered as a formal resignation.

If your employer puts a rule you have no way but to follow it.
However, to explain my contention...
Emails can be very easily falsified. Trail mails can be falsified even more easily. So it holds no evidential value. For forensic, the original mails is needed. It carries a certain amount of cryptic data including the server serial number against which it can be verified who actually sent the mail, when and how.
After you leave your account will be locked, so you can't get the original mail to prove it was sent, or by you or how. Someone else can be using your account back dating the mails and setting to create evidence against you.
